Situation: enemy squad is adjacent and in the open and makes a successful smoke attempt in its own hex.

This costs 1 MP

Question. Does this count a movement in the open for defensive fire purposes, does the smoke modifier apply or is it not considered movement and thus the unit cant be shot at until the defensive fire phase?

Good question. Can't find a rules reference for you, but I think you place the smoke before you're first fired on. That means you get the +2 Infantry Smoke Hindrance applied to the shot, which will negate FFMO. If you had declared Assault Movement to spend the 1MF to place the smoke, you'd also avoid FFNAM.

I'll be watching with interest the rules references that someone will no doubt be able to find! :)

I would think that, using the same logic of a unit is shot at 'after using x MF to enter a hex', so would a unit be shot at after placing the smoke

And it is an expenditure of MF, so the unit would definetly be a valid target for fire in the MPh

Seems to be a sequence question. Once a smoke attempt is declared are the 1MF (same hex) or 2MF (adjacent hex) expended then, which would allow the enemy to first fire without the +2 smoke modifier, or is the 1MF or 2MF spent AFTER the actual attempt?

Yes. With some exceptions (e.g. placement of a DEMO) all DFF must wait for an action to be completed. In the same way you cannot fire at a unit recovering a weapon and break that unit before it gets to roll for recovery.
