Do the LOS of an Observation Plane must be traced along an Hex Grain to the target (froma a FBE hex) for sighting TC purpose?

No. An observation plane makes sighting task checks per E7.61 "from any Friendly Board Edge hex of the owner's choice." It need not be four hexes away and it need not be down a hex grain.
